Kerstin and Sevastyan completed the assignment of H4-tail

H4-tail assignment

Kerstin Kaempf and Sevastyan Rabdano have completed the assignment of backbone 1H, 15N and 13C resonances. It was done using standard suite of triple resonance experiments (HNCO, HNcaCO, HNCA, HNcoCA, CBCANH and CBCAcoNH).

Nikolai and Kerstin took part in the international symposium “Magnetic Resonance: from fundamental research to practical applications” in Kazan, 21-23 Apr 2016.

Kerstin presented a poster entitled “Local and global dynamics of disordered proteins via the combination of temperature-dependent NMR spin relaxation measurements and MD simulations: a case study of H4 histone tail”. For the poster presentation she was awarded with one of the poster prizes.

Nikolai presented talk “Dynamics in protein crystals: insights from MD simulations complement new ssNMR and x-ray data”.

Biophysical Society 60th annual meeting

Sevastyan Rabdano and Sergei Izmailov attended Biophysical Society 60th annual meeting, Los Angeles, California, USA. They presented results of RRM2 and Guanylin oxidative folding projects.

Sevastyan presented poster “Loss of protein stability due to formation of intermolecular disulfide bonds under the effect of oxidative stress: case study of the RRM2 domain from neuropathological protein TDP-43”.

Sergei presented poster “MD Modeling of Oxidative Folding in Peptides and Proteins”.

The RRM2 mutant C198S was expressed and purified

Second mutant RRM2 construct was expressed and purified. The NMR studies of role of second cysteine in RRM2 will be performed.

The practical course “Cloning strategies”

Sevastyan Rabdano and Yulia Pivovarova attended two-week course on cloning in Moscow State University organized by Anna Golovina in Olga Dontsova’s lab (A.N. Belozersky Institute of Physico-Chemical Biology MSU). They were trained to perform cloning of DNA constructs to expression vectors including such operations like DNA electrophoresis, DNA extraction, PCR, restriction analysis.

Sevastyan presented the oral talk about projects in Biomolecular NMR laboratory at weekly group seminar.

Spinus 2015 conference

Sevastyan have attended the Spinus “Magnetic resonance and its applications” conference in Repino, St. Petersburg. The oral talk entitled “Последствия окислительного стресса для структуры и стабильности РНК-распознающего домена RRM2 белка TDP-43” was presented.

Sevastyan received main prize for best oral talk funded by Uwe and Barabara Eichhoff.

Finally the H4-tail has been successful expressed and now the first HSQC spectrum of this peptide has been recorded. Now the measurements of the dynamics can start!

The RRM2 mutant C244S was expressed and purified

This RRM2 mutant lacks cysteine that is presumably oxidized when wild-type RRM2 is treated by hydrogen peroxide. Experiment on this protein will help to understand the role of C244.
